چکیده

The planar cell polarity (PCP) signaling pathway is essential for a wide variety of celland tissue-level morphogenetic events in developing embryos. PCP research has been predominantly directed at the role played by a small number of core proteins, including Dishevelled and Frizzled. Much less is known about how these proteins signal downstream to direct the execution of such diverse cell behaviors as convergent extension and ciliogenesis. We have focused recently on the downstream PCP effector proteins, Fuz and Fritz. We show that Fuz is essential for ciliogenesis, but plays only a minor role in convergent extension. Combining bioinformatics with confocal and electron microscopy, we establish a central role for Fuz in membrane trafficking, showing that Fuz is essential for apical trafficking of ciliogenesis factors in ciliated cells and for exocytosis in secretory cells. Unlike Fuz, Fritz plays a major role in convergent extension. Time-lapse imaging reveals a cytoskeletal role for Fritz that is essential for the maintenance plasma membrane stability and cell shape. Together, these results provide novel insights into the mechanisms by which upstream developmental regulatory systems like PCP signaling are linked to the basic cellular machinery during embryonic morphogenesis.

Measurement of barbed ends, actin polymerization, and motility in live carcinoma cells after growth factor stimulation.

Motility is associated with the ability to extend F-actin-rich protrusions and depends on free barbed ends as new actin polymerization sites. To understand the function and regulation of different proteins involved in the process of generating barbed ends, e.g., cofilin and Arp2/3, fixed cell approaches have been used to determine the relative barbed end concentration in cells. The major disadv...

Cortactin is necessary for E-cadherin–mediated contact formation and actin reorganization

Classical cadherin adhesion molecules are key determinants of cell-cell recognition during development and in post-embryonic life. A decisive step in productive cadherin-based recognition is the conversion of nascent adhesions into stable zones of contact. It is increasingly clear that such contact zone extension entails active cooperation between cadherin adhesion and the force-generating capa...

In vitro behavior of silk fibroin-coated calcium magnesium silicate scaffolds

Bioceramic scaffolds such as silicate bioceramics have been widely used for bone tissue engineering. However, their high degradation rate, low mechanical strength and surface instability are main challenges compromising their bioactivity and cytocompatibility which further negatively affect the cell growth and attachment. In this study, we have investigated the effects of silk fibroin coating o...

Live Imaging of Cell Motility and Actin Cytoskeleton of Individual Neurons and Neural Crest Cells in Zebrafish Embryos

The zebrafish is an ideal model for imaging cell behaviors during development in vivo. Zebrafish embryos are externally fertilized and thus easily accessible at all stages of development. Moreover, their optical clarity allows high resolution imaging of cell and molecular dynamics in the natural environment of the intact embryo. We are using a live imaging approach to analyze cell behaviors dur...
